/Terminology / /Terminologie Bulletin of the World Health Organization, 64 (5): 767-770 (1986) © World Health Organization 1986 Allergen nomenclature* This article presents a nomenclature system for allergens which has been officially recommended by the International Union ofImmunological Societies (IUIS). The nomen- clature is based on proposals of the IUIS Sub-Committee for Allergen Nomenclature and is applicable to highly purified, well-characterized allergens and to non-purified or partially purified allergenic extracts. During the past several years, many antigens (aller- gens) which cause IgE-mediated atopic allergies in humans have been isolated from a wide variety of dif- ferent sources. The number of nomenclature systems associated with these allergens has grown almost in proportion to the number of newly described components. In order to bring some order into the present, chaotic situation, the IUIS Sub-Committee for Allergen Nomenclature has proposed a new unified nomenclature system for (a) highly purified allergens and (b) individual components identified within complex allergenic mixtures by means of various immunochemical and physicochemical separ- ation techniques. In the case of the highly purified allergens, the Sub-Committee also proposed guide- lines for the criteria used to establish homogeneity, envisaging that the specialities of allergen chemistry in particular and of human immune response in general will benefit substantially through the exchange of reagents and data. Comparable practical benefits should also accrue in the area of allergen standardization. Where necessary, a physical description of the source material (e.g., fungal spores, 95% purity) and any processing of the source material will be included. Highly purified allergens will be designated as follows: the first three letters of the genus (italicized); space; first letter of the species name (italicized); space; a Roman numeral. For example, Lol p I will refer to Lolium perenne allergen number one (perennial rye Group I (Rye I)). In the rare event that two species names have identical designations, they will be discriminated from one another by adding one or more letters (as necessary) to each species designation. The allergens will usually be numbered in the order in which they were isolated, or by general agreement of the IUIS Nomenclature Sub-Committee. The following general exceptions apply: (1) A major allergen will normally be assigned number one (e.g., Amb a I, short ragweed AgE; Gad c I, cod allergen M). (2) Structurally homologous (but not necessarily cross-reactive) components from different species will be assigned the same numbers (e.g., Der p I, Dermatophagoides pteronyssinus Pi and Derf I, D. farinae Agl 1). This may require numbering cer- tain allergens out of the sequence in which they were isolated or leaving gaps in the sequence of the numbers of allergens from certain species (e.g., as in the case of Ambrosia trifida, giant ragweed (Table 1)). It is also recognized that in exceptional cases, where homology is established after the initial nomenclature has been assigned, some revision of the 4722 -767 ALLERGEN NOMENCLATURE Table 1. (This may possibly be required in the case of the Phleum pratense (timothy) allergens). (3) Homologous, immunologically closely related components within the same species (isoallergens) will be designated by capital letters (A,B,C, etc.) in have been described in this species. the order of decreasing pl (e.g., Amb a VA, Ra5A (pl 9.6) and Amb a VB, Ra5B (pl 8.5)). If two closely related components have similar pl values (differing by less than 0.05 pl unit), they will be designated in the order of decreasing relative molecular mass, as determined by SDS-PAGE (sodium dodecyl sulfate 768 ALLERGEN NOMENCLATURE 769 polyacrylamide gel electrophoresis) under reducing conditions,a or by more rigorous procedures such as sequence analysis (if not differentiated by SDS- PAGE). The Sub-Committee recognizes that a few aller- gens, e.g., Gal d I (ovomucoid) and Poa p X (cyto- chrome c), already have been assigned biochemical names. In such cases, both the assigned allergen nomenclature and the biochemical name should first be specified and may be used interchangeably thereafter. In the case of fungal allergens, where there is already evidence for strain differences within a species, the American Type Culture Collection (ATCC) number (or other appropriate numerical designation) will be included as a subscript after the allergen number, where it is necessary to designate such differences (e.g., Alt a I63, Alt-I isolated from Alternaria alternata, ATCC No. 6663, which is slightly different antigenically from Alt a I16086). It is envisaged that a similar approach will be used in the future when strain or subspecies differences identi- fied in other biological sources are described. In regard to nomenclature, no attempt will be made to discriminate between components of different parts of the plant or animal source, nor between allergens that enter the body by different routes. The Sub-Committee appreciates that relatively clearly defined differences exist with respect to the route of exposure in certain cases (e.g., chicken eggs versus feathers), but in other cases (e.g., fish proteins) the route of sensitization may be by ingestion or inhalation, or by both of these routes. Further abbreviated usage. Where reference is made to an allergen from a particular, clearly desig- nated species (e.g., within a publication dealing with a single specified allergen source), the inclusion of the last italicized letter for the species will not be neces- sary (e.g., Fel I, cat allergen 1). Purity requirements. In order to be designated according to the new system, the allergen must possess a high degree of purity according to several selected physicochemical and immunochemical cri- teria. At least four and preferably five different types of criteria are recommended, including (but not limited to) appropriate selection of separation tech- niques based on the following properties: ' Protein allergens having non-identical polypeptide chains which are held together by disulfide bonds and/or by non-covalent bonds, give multiple bands on SDS-PAGE under reducing con- ditions. In such cases, the relative molecular mass of a protein shall be the sum of its polypeptide chains required to form the monomeric protein unit as established by gel filtration or gel electrophoresis in the absence of denaturant. Molecular size: SDS-PAGE, gel filtration. Molecular charge: Isoelectric focusing (IEF), elec- trophoresis (PAGE, agarose gel, starch gel, etc.), ion-exchange chromatography (especially high- performance liquid chroma- tography (HPLC) using an ap- propriate anionic or cationic exchanger). Immunochemical: Crossed immunoelectrophor- esis/crossed radioimmunoelec- trophoresis (CIE/CRIE) or immunoelectrophoresis (IEP) using hyperimmune antisera from at least three outbred animals. Hydrophobicity: Reverse-phase HPLC. Chemical: NH2-terminal amino acid deter- mination, COOH-terminal amino acid determination, amino acid composition. In each case, the experimental conditions should be clearly defined (see also footnote a). It is recognized that closely related isoallergens are often not readily separable. In the case of iso- allergenic mixtures, the pI and/or molecular size range will be specified. Other requirements. The following physical and chemical parameters will be supplied where possible: relative molecular mass (under defined conditions; see also footnote a), amino acid composition and sequence, carbohydrate content and composition (as well as location and type of linkage if known), extinction coefficient (including conditions for measurement), nitrogen content, presence of pros- thetic groups, enzymatic or other biological activity (if any), and X-ray crystallographic structure. Where appropriate international (and/or national) reference preparations are available, such as those produced through the IUIS-WHO allergen standardization programme, the investigator should provide a quanti- tative estimate of the content of the purified allergen in the reference; furthermore, in-house crude allergen preparations should also be standardized against the reference. Each investigator should attempt to define the allergenic importance of the isolated allergen in a sub- population of subjects who possess IgE antibody and/or react positively by skin test to a well-charac- terized, standardized extract or the crude allergenic mixture from which the purified allergen was derived. The .allergic population studied should be clearlydefined epidemiologically. In addition, the method ALLERGEN NOMENCLATURE of in-vivo and/or in-vitro testing used, and the criteria for positivity and quantitation of the degree of responsiveness, should be described. The Sub-Committee further proposes that investi- gators should send their data and publications on highly purified allergens to the Chairman (Dr D. G. Marsh) in order that they may be made available to other investigators, and be periodically updated. It is envisaged that the availability of such a compre- hensive record will be beneficial to researchers in many areas of human immune response and of atopic disease in particular. Finally, and most importantly, the Sub-Committee recommends that each investigator reporting the purification of an allergen should, as a minimum requirement, be willing to supply a sufficient amount of a monospecific antibody (polyclonal or mono- clonal) to selected qualified investigators in order to allow them to identify the allergen immuno- chemically. Non-purified or partially purified allergenic extracts CIE/CRIE and related technologies. Antigens (Ag) will be designated by Arabic numerals, starting with those showing greatest anodic mobility at pH 8.6 (e.g., Cla h Agl, Ag2, etc.). Researchers should take care in defining the antisera used in CIE/CRIE and be willing to make the sera available to other labora- tories in order to facilitate inter-laboratory com- parisons. IEF "immunoprinting" and related technologies where the pI is defined. Each IEF band (Bd) will be numbered according to its pI, rounded to the nearest 0.05 pIunit (e.g., Dac g Bd5.9). SDS-PAGE and related technologies where the relative molecular mass is defined. Each band will be designated according to its relative molecular mass (e.g., Derf Bd17K), where the abbreviation "K" refers to "thousand". Otherpoints. The terminology used for CIE/CRIE will be kept distinct for that application. In the case of IEF and SDS-PAGE, where both the pI and relative molecular mass are known, the corres- ponding band may be designated using both types of data e.g., Derf Bd3.75/30K. The genus and/or species abbreviations may be omitted in a situation where there is no confusion concerning designation and where the species has been previously defined. The Nomenclature Sub-Committee encourages collaborative efforts among investigators working with the same or a related species to exchange infor- mation and reagents in order to minimize confusing nomnnclature differences between laboratories. The laboratory reference patterns obtained using the investigator's own reagents should be compared with those obtained using appropriate international or national reference preparations, where available. Highly purified allergens, covered under the standard nomenclature system, should also be identified in the reference patterns by the investigator, wherever possible. The Sub-Committee recommends that all defined, but not fully purified components (e.g., antigens recognized by CIE) should be investigated for allergenic activity (e.g., by CRIE) using at least 20-30 human sera from individuals who have high levels of IgE antibodies to the crude allergenic extract containing the defined component. Attempts should also be made to identify antigenically and aller- genically cross-reactive components. IUIS Sub-Committee for Allergen Nomenclature David G. On admet aussi que dans des cas exceptionnels, oiu l'homologie est demontr6e apres 'tablissement de la nomenclature initiale, une revision de la nomenclature peut s'imposer. (Tel sera peut-etre le cas des allergenes de Phleum pratense (fleole)). 3) Des composes homologues, etroitement appa- rentes du point de vue immunologique, au sein de la meme espece (isoallergknes) seront designes par des lettres capitales (A, B, C, etc.) par ordre de pI de- croissant (par exemple Amb a VA, Ra5A (pl 9,6) et Amb a VB, Ra5B (pI 8,5)). Le Sous-Comite n'ignore pas que quelques aller- genes, par exemple Gal d I (ovomucoide) et Poa p X (cytochrome c), ont deja recu un nom biochimique. Dans ce cas, on precisera d'abord le nom etabli selon la nomenclature des allergenes et le nom bio- chimique, puis on les utilisera indifferemment par la suite. Dans le cas des allergenes fongiques, lorsqu'il existe deja differentes souches au sein d'une espece, on indiquera en indice, apres le numero de l'aller- gene, le numero de l'American Type Culture Collec- tion (ATCC) (ou toute autre designation num6rique appropriee), lorsqu'il est n6cessaire de mentionner une telle difference (par exemple, Alt a I6"3, c'est-i- dire Alt-I isole d'Alternaria alternata, N° ATCC 6663, qui est l6gerement different du point de vue antig6nique d'Alt a I16086). On envisage d'adopter une approche analogue lors de la description de sources animales ou vegetales pour lesquelles il existe des differences de sous-especes ou de souches. En ce qui concerne la nomenclature, on ne cher- chera pas A distinguer les constituants des diff6rentes parties de la plante ou de l'animal, ni les allergenes qui penetrent dans l'organisme par des voies diff& rentes. Le Comite est toutefois conscient qu'il existe des differences assez claires en ce qui concerne la voie d'exposition dans certains cas (par exemple, pour la poule, les aeufs ou les plumes) mais dans d'autres cas (par exemple, les proteines des poissons) la sensibili- sation peut se faire par ingestion ou par inhalation, ou encore par ces deux voies associees. Designation abrigge. Lorsqu'on parle d'un aller- gene appartenant a une espece definie, clairement designee (par exemple, dans une publication traitant d'une source unique d'allergene, clairement sp6ci- fi6e), il n'est pas necessaire de mentionner la lettre en italique de l'espece (par exemple, Fel I, allergene de chat 1). Criteres de purete'. Pour recevoir une designation conformement au nouveau systeme, I'allergene doit etre d'une tres grande purete, etablie selon plusieurs criteres physicochimiques et immunochimiques. Immunochimie: immunoelectrophorese croisee/ radioimmunoelectrophorese croisee (CIE/CRIE) ou IEF avec des serums hyperimmuns d'au moins trois animaux non consanguins. Hydrophobie: HPLC a phases invers6es. Chimie: determination des acides amines NH2-terminaux et COOH-ter- minaux, composition en acides amines. Dans chaque cas, les conditions experimentales seront clairement indiqu&es (voir egalement note a). On sait que les isoallergenes etroitement apparen- t6s sont souvent difficiles a separer. Dans le cas de melanges d'isoallergenes, on pr6cisera le pl et/ou la gamme de masses moleculaires. Autres crit0res. On indiquera si possible les para- metres physiques et chimiques suivants: masse mol- culaire relative (dans des conditions bien definies; voir egalement note a), composition en acides amines et sequence d'acides amines; teneur et composition en glucides (avec localisation et type des liaisons, s'ils sont connus), coefficient d'extinction (avec indi- cation des conditions de mesure), teneur en azote, pr6sence de groupements prosthetiques, activite enzy- matique ou autre activite biologique (le cas echeant) et structure cristallographique d6termin6e aux rayons X. Lorsqu'il existe des preparations internationales (et/ou nationales) de reference appropriees, telles que celles produites dans le cadre du programme UISI-OMS de standardisation des allergenes, on donnera une estimation quantitative de la teneur de la preparation de reference en allergene purifie; de plus, les pr6parations brutes d'allergenes utilis6es en laboratoire devront egalement etre standardis6es par rapport a la preparation de reference. Les profils de ref6rence obtenus au laboratoire avec les r6actifs d'un chercheur devront etre compares avec ceux obtenus au moyen de preparations inter- nationales ou nationales de reference appropriees, si elles existent. Les allergenes hautement purifies couverts par le systeme normalise de nomenclature doivent aussi etre identifies par le chercheur selon leur profil de reference. Le Sous-Comite recommande que tous les constituants definis, mais non totalement purifies (par exemple les antigenes reconnus par CIE) soient etudies du point de vue de leur activite allerge- nique (par exemple par CRIE), en utilisant au moins 20 a 30 serums de sujets pr6sentant un taux eleve d'IgE a l'egard de l'extrait allergenique brut qui con- tient le constituant en cause.
