EXPANDED PROGRAMME ON IMMUNIZATION The Effects of Freezing on the Appearance, Potency and Toxicity of Adsorbed and Unadsorbed DPT Vaccines The effects of freezing on adsorbed DPT vaccines were reported in the previous issue of the WER. In the present issue the results obtained with adsorbed D T and unadsorbed D PT vaccines are shown. Adsorbed DT Vaccines (Table 2) Adsorbed DT vaccines underwent similar physical changes after being frozen as adsorbed DPT vaccines from the same laboratory. Significant loss of potency was observed in the tetanus toxoid com­ ponent after freezing at —30°C. At —5°C, the reduction in potency was not significant in one lot tested, while a second lot decreased to 61 % of the potency of the unfrozen control. Although the potency of the tetanus component was substantially reduced after freezing, the remaining potency still exceeded WHO requirements in this respect (40IU per single human dose). Previous experience of the laboratory revealed that the potency of both diphtheria and tetanus components of the DT vaccine declined by more than 75% after freezing and thawing it 6 times.

Dans ce numéro on trouvera les résultats obtenus avec les vaccins D T adsorbés et DTCoq non adsorbés. Vaccins DT adsorbés (Tableau 2) Les vaccins DT adsorbés subirent les mêmes modifications physiques après congélation que les vaccins DTCoq adsorbés du même laboratoire. Une diminution sensible de l’activité de l’ana­ toxine tétanique fut observée après congélation à —30°C. A —5°C, la diminution d ’activité n ’était pas sensible dans un des lots examinés tandis que dans l’autre l’activité se trouva ramenée à 61 % de celle de l’échantillon témoin. L ’activité de l’élément antitétanique, bien que sensiblement réduite après congélation, dépassait tout de même les prescriptions de l’OMS à cet égard (40 UI par dose unique pour l’homme). L ’expérience passée du laboratoire indique que l’activité des éléments antidiphtérique et antitétanique du vaccin DT diminue de plus de 75% après six séries de congélation et de décongélation.

Granular appearance following freezing at —5°C and —30°C for 12 hours. This effect was just detectable with naked eye when the samples were viewed in a strong light and was associated with a reduction in opacity of the material when compared with the control samples stored at 4- 4°C. With the aid of a hand lens the particulate matter in the vaccine could be readily seen to have aggregated. Aspect granuleux après congéla­ tion pendant 12 heures à —5°C et —30°C. Unadsorbed DPT Vaccines (Table 3) Among the three unadsorbed DPT vaccines tested, none showed evidence of physical changes after being frozen. However, after six cycles of slow freezing at —18°C and thawing, carried out in one laboratory (No. 9), vaccines became heterogenous and exhibited a finely coagulated and somewhat fibrous appearance, compared with the uniform turbidity of the controls. All vaccines were tested for pertussis potency and it was reduced in one. Two vaccines were also tested for tetanus toxoid potency, which was not reduced. One of the vaccines was tested for diphtheria potency, which was not reduced. The toxicity of the pertussis component of all three vaccines tested remained unchanged after single or repeated freezing.

Cependant, après six séries de congélation lente à —18°C et de décongélation, effectuées dans un des laboratoires (N° 9), les vaccins étaient devenus hétérogènes, présentant un aspect finement coagulé et quelque peu fibreux, par rapport au trouble uniforme des échantillons témoinsL'activité de l'élément anticoquelucheux fut contrôlée dans tous les vaccins; elle était réduite dans un. L ’activité de l’anatoxine téta­ nique fut aussi contrôlée dans deux vaccins; elle n ’était pas réduiteL’activité de l’élément antidiphtérique, contrôlée dans un vaccin, n ’était pas réduite. La toxicité de l’élément anticoquelucheux des trois vaccins examinés demeura inchangée après une ou plusieurs congélations.

Samples frozen once remained uniformly opalescent and no clear-cut differences between samples frozen at — 15°C and — 30°C and control samples kept a t + 4 “C were observed neither in regard to the am ount o r type o f deposit, n o r the degreee o f opa­ lescence o f the residual suspen­ sion. N o evidence for aggrega­ tion or lysis o f B, pertussis cells during freezing and thawing has been obtained.
